Exploring Polyploid Induction in Lilium Using Chemical Agents in Tissue Culture

Abstract

The potential for inducing polyploidy in Lilium species is substantial for enhancing ornamental traits and genetic diversity. To examine the efficacy of colchicine, oryzalin, and caffeine in stimulating polyploidy through bulb-scale tissue culture, this study was conducted. Varying concentrations of these chemical agents were employed to assess their impact on polyploid induction. Concentrations of colchicine (0.05%, 0.1%, 0.2%), oryzalin (0.001%, 0.003%, 0.005%), and caffeine (0.6%, 0.9%, 1.2%) were applied for 3 hours, followed by washing with deionized water and streaking onto half MS medium. The half-MS medium consisted of 30 g sucrose, 2.2 g MS, 4 g gelrite, and a pH of 5.8. Observations were made in the dark culture. Interestingly, colchicine treatment resulted in lethality, suggesting potential challenges with this method. However, the use of oryzalin (0.001%, 0.003%) and caffeine (0.6%, 0.9%) at specific concentrations yielded promising results, successfully stimulating callus differentiation. These findings shed light on the complex mechanisms underlying polyploid induction in Lilium and pave the way for refining protocols to optimize polyploid breeding strategies in ornamental plants. Polyploidy induction enables the rapid generation of genetic variation to develop new traits and characteristics. Further cytologic studies will be conducted to confirm the polyploidy.
