Analysis of Growth Potential of Single-Stemmed Rose ‘Antique Curl’ by Node Cutting Position
삽수 채취 절위에 따른 절화장미 ‘엔틱컬’ 단경삽목묘의 생장 활력 분석

Abstract


We investigated the growth potential of cuttings by node position in order to find the optimal node for single-stemmed roses ‘Antique Curl’ with the goal of increasing the mass-production efficiency of cut rose cuttings. Single-node cuttings from six nodes (six levels) with five leaflets below the peduncle were grown in a greenhouse and environmentally controlled growth chambers for one month. The lower the node cutting came from, the more the rooting and bud-break of that cutting was affected by early leaf etiolation and falling. The initial carbohydrate content of the leaves did not affect rooting and bud-break rate. Levels 1 – 3 in the greenhouse and levels 1 – 2 in the environment-controlled growth chamber were the best cutting positions for rooting and bud-break, and nodes up to level 4 showed the expected growth of roots and shoots. The node positions up to level 4 would therefore be available for mass cutting propagation in commercial greenhouse rose production.




절화장미 삽목묘 대량생산 효율을 높이기 위해 국내 육성 절화장미 ‘엔틱컬’(Rosa hybrida ‘Antique Curl’)을 대상으로 생 장 활력이 높은 단경삽목묘 주요 절위를 분석하였다. 화경으 로부터 순서대로 5매엽 이상을 가진 마디 6개를 삽목(6 절위, Level 1~6)하여 온실과 생장환경조절실에서 각각 30일간 관리 하였다. 그 결과, 삽목 절위가 기부 쪽으로 내려갈수록 발근율 과 액아 생장이 억제되었고, 삽수 잎의 황화 및 낙엽 현상이 조기에 나타났다. 잎의 탄수화물 함량은 발근과 신초발아율에 영향을 주지 않았다. 온실환경에서 1~3 절위, 생장환경조절실 에서 1~2 절위의 생장이 가장 우수하였고, 모두 4절위까지는 정상적인 생장을 보였다. 결과적으로 상업용 장미 시설 생산 에서 대량삽목번식을 할 때 화경으로부터 5매엽을 가진 네번 째 마디(level 4)까지 가능할 것으로 확인되었다.



초록


    서 언

    장미는 국화, 카네이션, 튤립과 함께 세계 4대 절화이며 화 색, 향기, 화형 등의 다양한 특성을 가지고 있어 국내 화훼시 장에서도 소비자 선호도가 매우 높다(Yeon and Kim 2020a;Yeon and Kim 2020b). 절화용 장미는 삽목, 접목, 접삽목, 아 접 및 조직배양 등 다양한 방식으로 번식되지만, 그 중 삽목 번식은 작업이 간편하고 생산비용이 적어 대량생산에 용이하 다(Jeong et al. 2007). 특히 한 마디를 잘라 번식하는 단경삽 목(single node cutting)은 생육 초기 신초 생장과 개화를 촉 진하여 상품성 절화 수확에 필요한 기간을 단축할 수 있어 절 화장미 생산 효율을 증진시킬 수 있다(Bredmose and Hansen 1996). 하지만 단경삽목묘는 절화장미 생산을 위한 생체량이 제한되어 있기 때문에 초기 생장 활력이 높은 것이 유리하다 (Kim 2009). Hartmann et al.(1996)는 삽목묘 발근 정도는 화 학발근제 종류 및 처리 수준, 발근 환경, 모체(mother shoots) 의 생리적 상태에 의해 영향을 받는다고 보고하였다. 특히 mother shoots의 생리적 요인 중 품종, 연령, 그리고 삽수 채 취 부위(node position)가 삽목묘의 발근 속도와 생장 양상의 차이를 유도하는 것으로 알려져 있다(Bredmose and Hansen 1996;Kim 2009;Park et al. 2011).

    따라서 본 연구에서는 국내 화훼시장에서의 선호도가 높은 복색 꽃잎을 가진 국내 육성 스탠다드형 절화 장미 엔틱컬 품 종(‘Antique Curl’, 농촌진흥청 2011년도 육성)을 대상으로 초 기 생장 활력이 높은 단경삽목묘 확보를 위해 삽수 채취 마디 별 발근과 신초 생장 특성을 분석하였다.

    재료 및 방법

    식물 재료

    경기도 파주시 소재 장미 재배온실에서 ‘엔틱컬’(Rosa hybrida ‘Antique Curl’)을 수확하여 농가 현장에서 단경삽목용 삽수를 조제하였다. 삽수는 소엽수가 5장 이상인 마디(5매엽 마디)를 대상으로 하였고 일괄적으로 소엽수를 4장으로 정리하였다. 삽수 채취 마디 위치에 따른 생장 활력 검정을 위해 화경으로 부터 첫번째 5매엽 마디를 1절위(Level 1)로 구분하여 기부 방 향으로 차례로 6번째 마디까지 2-6 절위(Level 2, 3, 4, 5, 6)로 구분하였다. 삽수의 발근 능력 검정을 위하여 별도 호르몬 처 리는 진행하지 않았다. 삽수의 기초 생체정보를 알아보고자 삽목 전에 절위별로 삽수 길이와 무게를 측정하고(n=15), 잎 의 탄수화물 함량을 분석하였으며(n=5), 액아(axillary bud)를 검경하였다(IMS-M-345, Sometech Vision, Korea).

    잎의 탄수화물 함량

    절위 및 엽령에 따른 생장 포텐셜 차이를 분석하기 위해 각 절위별 잎의 탄수화물 함량을 측정하였다(Park et al. 2015). 고온건조 후 분쇄한 시료 0.05g에 0.7N HCl 10mL을 첨가해 중탕(100℃, 150분)한 후 미리 차갑게 만든 시험관에 원심분 리(5000rpm, 23℃, 30분)한 상등액 0.05mL와 증류수를 첨가 하여 1mL mass-up했다. 0.2% anthrone(Sigma, Slovakia)(95% H2SO4) 4mL을 첨가 후 재중탕(100℃, 8분)했다. 얼음 중탕하 여 발색이 멈춘 샘플을 흡광분광분석기(UV-2450, Shimadzu Corp., Japan)를 이용하여 파장 620nm에서 비색정량하였다. 검량선은 glucose(Sigma, USA)를 이용한 표준용액으로 작성 하였다.

    환경별 삽목묘 발달 차이

    환경에 따른 절위별 단경삽목묘의 발달 차이를 알아보고자 서울시립대학교 실험용 유리온실과 생장환경조절실(HB-301S-3, Hanbaek Scientific Co., Korea)로 구분하여 2020년 7월 1일 삽목 진행했으며 7월 30일까지 30일 동안 Table 1과 같이 관 리하였다. 온실 재배 시, 실험 3반복과 개체 10반복(n=30), 생 장환경조절실 재배 시, 개체 15반복을 진행했다. 이후 삽목묘 절위별 잎의 황화, 뿌리와 신초 발생 특성, 신초 길이, 건물중 을 측정하였다. 잎의 황화 현상은 노란 반점 발생을 기준으로 측정하였고, 줄기 부패 시 폐기하였다. 발근율과 뿌리 길이는 가장 긴 뿌리(>1mm)를 기준으로 조사하였다.

    통계분석

    통계분석 프로그램인 SAS 9.4(SAS Institute, Cray, NC, USA) 를 이용하여 ANOVA 분석(analysis of variance), student’s t-test, DMRT(Ducan’s multiple range test)를 진행하였으며 유의 확 률은 5% 수준으로 설정하였다.

    결과 및 고찰

    절위별 삽수 생장포텐셜

    모주에서 형성된 절위별 액아의 크기는 1 절위가 가장 컸으 며 하위 마디인 6 절위로 갈수록 감소하였다(Fig. 1). ‘엔틱 컬’ 삽수의 줄기 직경은 절위가 기부로 내려갈수록 커졌으나 (Table 2), 삽수의 절위별 마디길이(Table 2), 생체중, 건물중, 엽면적(Table 3), 잎의 단위 무게당 탄수화물 함량(Fig 3B)은 유의미한 차이를 보이지 않았다. 탄수화물은 식물이 자체적으 로 광합성을 하여 축적한 동화산물로, 식물 발달에 에너지원 이자 골격요소로 사용된다(Corrêa et al. 2004). 또한 자당은 절화장미를 비롯한 여러 식물 액아의 신초 발생을 조절한다 (Kebrom 2017). 잎의 탄소 동화능력은 엽령이 높을수록 감소 하고(Gonzalez-Real and Baille 2000) 엽령은 줄기 기부에서부 터 화경쪽으로 올라갈수록 낮아진다(Ma et al. 2015). 하지만 ‘엔틱컬’ 잎의 총 건물중(Table 3)과 단위무게당 탄수화물 함 량(Fig. 3B)은 엽령에 따른 뚜렷한 차이가 나타나지 않았기 때 문에 잎의 탄수화물 함량이 단경삽목묘의 절위별 생장 차이를 야기하는 주요인은 아니라고 판단한다. 따라서 개화줄기에서 함량이 높은 비구조성 탄수화물(Mattson et al. 2008)과 생육 할 때 동화된 탄수화물이 삽목묘의 발달에 더 영향을 주었을 것으로 생각할 수 있다.

    ‘엔틱컬’ 삽목묘의 폐기율과 잎의 황화율은 근권부 방향의 절위일수록 높아졌다(Table 4). 장미 잎의 황화 및 부패 현상 은 수분 스트레스나 질소, 칼륨, 마그네슘과 같은 다량 영양 소 결핍으로 나타날 수 있고, 특히 질소와 칼륨은 식물체 내 이동성이 높아 결핍 현상이 보다 더 오래된 잎에서 발생한다 (Hopkins and Hüner 2009; Lee et al. 2016). 또한 잎의 칼 륨과 질소 함량은 기부로 갈수록 감소하며 상위 잎에 의한 간섭으로 질소함량기울기가 발생한다(Hopkins and Huner 2009). 따라서 동일 환경조건에서 황화 및 부패율이 아래 절 위에서 높았던 것은 하부 절위의 다량원소 부족이 원인인 것 으로 판단하였다.

    환경에 따른 절위별 신초 생장 특성

    온실환경에서 ‘엔틱컬’ 삽수는 1 절위부터 5 절위까지 발 근율이 70% 이상이었고 5 절위부터 뿌리길이가 감소하였다 (Table 5, Fig. 2). 6 절위에서는 발근율과 뿌리길이 및 건물중 이 다른 절위 삽수보다 매우 낮았다. 신초 발아는 1~4 절위에 서 발생하였고 생장량은 2 절위에서 가장 크며 절위가 기부로 갈수록 감소하는 경향이 있었다. 2 절위의 신초 생장량이 가 장 높은 것은 유리온실에서 재배된 ‘Sonia’ 결과와 유사하였다 (Bredmose and Hansen 1996).

    생장환경조절실에서 자란 ‘엔틱컬’ 삽수의 발근율은 1~4 절 위에서 80% 이상이었으며 5 절위부터 급격하게 감소하였다 (Table 5, Fig. 2). 뿌리 길이 및 건물중은 1 절위에서 가장 크 며 기부로 내려갈 수록 감소하는 경향이 있었다. 생장 한 달 동안 1~4 절위에서 신초가 발생하였고, 1 절위에서 가장 큰 생장량이 나타났으며 기부로 내려갈수록 감소했다. 이런한 결 과는 Rosa hybrida ‘Ruidriko’와 ‘Teresa’등의 결과와 유사하였 지만(Le Bris et al. 1998;Kim 2009), Dieleman et al. (1997) 가 보고한 기부 채취 삽수 생장에 미치는 사이토키닌의 긍정 적 효과와 상이했다. 정단에 가까운 삽수 일수록 신초 및 뿌 리 발생율이 높았던 이유는 모주의 기부 마디가 상대적으로 일찍 형성되어 액아의 생리 활력 차이를 나타냈으며(Fig. 1) 오옥신의 활성과 연관이 있는 것으로 해석된다(Kim 2009)

    온실과 생장환경조절실 두 환경에서 재배한 삽수의 뿌리와 신초 생장량은 절위 수준이 내려갈수록 뚜렷하게 감소했다 (Table 5). 환경조건에 따른 발근 및 신초 발생률 차이는 크지 않았지만(Fig. 4), 절위별 삽수 생장량은 큰 유의적 차이를 보 이며 상이했다(Table 5, Fig. 3A). 총 건물중으로 분석한 1~4 절위의 생장량이 생장환경조절실환경에서 온실보다 약 6.3 배 높게 나타났고(Fig. 3A), 이는 95% 차광 및 포그가 설치되어 온도와 상대습도가 삽수 생장에 적합한 범위로 유지되는 온실 내부 삽목상의 환경과 비교하여(Table 1) 생장환경조절실의 높은 광도(300μmol・m-2・s-1)가 뿌리와 신초의 생장을 촉진한 것으로 판단한다(Choi et al. 2000). 생장환경조절실에서 상대 적으로 신초 생장량이 높았던 것은 결과적으로 다량원소 사용 증가로 이어져 기존 잎의 높은 황화율을 야기한 것으로 판단 된다.

    결론적으로 절화장미 ‘엔틱컬’대상으로 상업적인 단경삽목 묘를 획득하고자 할 때 꽃으로부터 기부방향으로 5소엽을 가 진 1~4 절위가 적합한 것으로 확인되었고, 삽수의 발근 및 신 초 발아 포텐셜은 상위 절위에서 높았으며 이것은 삽수의 액 아 활력 수준과 잎의 영양상태에 따른 것으로 확인되었다.
