Intermediate Physiological Dormancy in Seeds of Prunus maackii Native to Korea

Abstract

The genus Prunus includes more than 200 species of trees and shrubs widely distributed in temperate zones and are valued for their ornamental features. Studies about seed dormancy type and germination requirements provide useful information for developing propagation protocol of P. maackii. The seeds of P. maackii were permeable to water and had a mature embryo during seed dispersal. In the temperature experiments, the seeds at 4, 15/6, 20/10 and 25/15°C (12/12, light/dark) had less than 10% germination within 20 weeks. The seeds treated with control, endocarp removal, endocarp removal + GA3, endocarp removal + fluridone, and surfuric acid scarification germinated to 0, 33, 81, 63, and 0%, respectively, at 25/15°C after 8 weeks of incubation. The seeds treated with GA3 of 0, 10, 100, 1000 mg・L-1 germinated to 0, 6, 3 and 50%, respectively, at 25/15°C after 20 weeks of incubation. The seeds treated with 0, 2, 4, 8, and 12 weeks of cold stratification (4°C) germinated to 0, 16, 19, and 16%, respectively. Therefore, the seeds are in a physiologically dormant state at suitable conditions. The endocarp of P. maackii has an important role of germination inhibitor. 12 weeks of cold treatment alone was not enough to break physiological dormancy, and GA3 was effective in promoting germination. Thus, the seeds of P. maackii have intermediate physiological seed dormancy.
