Application of Plant Cytogenetics in Floricultural Research
화훼연구에 있어서 식물세포유전학의 응용

Abstract

Cytogenetics, which is concerned with study of chromosome number, structure, and behavior, is derived from merger of cytology and genetics. Furthermore, molecular cytogenetics is a relatively recent development in this field by incorporating applications of molecular biology techniques and has gained popularity since the 1980s. With the start of whole genome sequencing projects, cytogenetics have been utilized in various research fields. Research tools in cytogenetics include banding techniques by conventional staining methods, fluorescence in situ hybridization (FISH), genomic in situ hybridization (GISH), fiber FISH, and BACFISH. These advanced technology in cytogenetics widen the scope of applications ranging from basic karyotype analysis to applied research like exploring the location of a gene. The use of cytogenetic studies in floricultural research provide: (1) a basic information of ploidy level in breeding programs, (2) precise karyotype analysis by FISH technique, (3) analysis of introgression of alien genome in interspecific hybrid by GISH technique, (4) genome analysis using micro-dissection technique, and (5) detection of single-copy genes in GM crops. However, cytogenetic research in local floriculture has lagged behind the global trend. In order to further genome studies in flowering plants it is necessary to utilize various techniques in a variety of crops. In this review paper, we discuss the concept and essence of cytogenetics in floricultural research.

    세포유전학과 염색체

    세포유전학(Cytogenetics)은 문자 그대로 세포의 유전 (Cell genetics)에 관해 연구하는 학문으로 세포학(Cell biology)과 유전학(Genetics)으로부터 형성된 학문이다. 최 근에는 분자생물학(Molecular biology)적 기술이 적용됨에 따라 학문의 이용범위와 연구의 내용이 보다 확대되어 분 자세포유전학(Molecular cytogenetics)라고 불리고 있다. 기 본적으로 세포유전학은 염색체의 수, 구조, 염색체 이 상(chromosomal aberration), 염색체 행동(chromosomal behavior), 체세포 및 감수분열 등의 염색체 연구를 기본 으로 하는 연구분야이다(Mason 2013).

    염색체는 1840년대 스위스의 식물학자 Nageil에 의해 식물세포 핵 안에 실과 같은 구조(tread-like structure)로 묘사되며 ‘transitory cytoblast’라고 명명되었으며 이후 1888년 Waldeyer가 염색이 잘되는 물질(chromos = Greek for color, soma = Greek for body)이라는 뜻의 염색체 (chromosome)라 명명되었다. 염색체의 형태는 동원체 (centromere)를 기준으로 길이가 짧은 부분을 단완(short arm 또는 p-arm), 길이가 긴 쪽을 장완(long arm 또는 q-arm)이라 명명한다. 동원체의 위치에 따른 염색체의 조 성은 Levan et al.(1964)의 기준에 의해 장완과 단완의 길이비(arm ratio)가 1.00에서 1.67 미만은 중부동원체형 (metacentric, m), 1.67 이상 3.00 미만을 차중부동원체형 (submetacentric, sm), 3.00 이상 7.00 미만을 차단부동원체형 (subtelocentric, st), 7.00 이상을 단부동원체형(telocentric, t)으 로 구분하고 있다.

    핵형(karyotype)은 하나의 핵 안에 존재하는 중기상 (metaphase stage)의 염색체의 수, 형태, 크기 등의 여러 특성을 조사하여 나타내는 것으로 염색체의 크기, 모양, 동원체의 위치에 의해 상동염색체를 구별하여 주로 길이의 감소함에 따라 나열하는 것을 핵형도(Karyogram)라고 한다.

    Gupta(2006)는 시대에 따라 세포유전학적 연구의 주요 내용을 다음과 같이 분류하였다. 첫번째 시대(1910 ~ 1970 년)에는 주로 하나의 세포 안에 포함된 염색체의 수를 조 사하는 염색체의 배수성(Ploidy) 및 이수성(aneuploidy) 연 구, interchange, inversion 등과 같은 염색체 구조 연구 가 주로 이루어졌다. 이와 같은 연구는 주로 Aceto- Carmine, Feulgen, Aceto-Orecein 등과 같은 전통염색법 을 이용하여 광학현미경하에서 관찰되었다. 또한 시대에 는 Giemsa-C banding technique이 개발되어 constitutive heterochromatin region에 특이한 패턴으로 나타나는 band 를 분석하는 C-banding 기법도 많이 이용되었다(Vosa and Marchi 1972). 그러나 식물염색체는 식물마다 염색 체의 숫자가 다양하고 그 크기가 비교적 작아 상동염색체 를 정확히 짝짓고 특징을 짓는 데는 한계가 있었다. 두번째 시대(1950 ~ 1980년) cytospectrophotometry와 rereassociation kinetics를 이용하여 haploid content(1C value)와 현화식물의 게놈에서 많은 부분을 차지하는 repetitive sequence의 구 성을 연구하기 시작하였다. 세번째 시대(1980 ~ 현재)에는 1980년대 후반에 염색체 상에 biotin이나 digoxygenin과 같은 형광물질로 표지된 hapten을 혼성화하는 in situ hybridization 방법이 개발되었다. 이러한 방법의 개발은 특정 DNA 염기서열의 위치를 염색체 상에서 밝혀냄으 로써 물리적 유전자지도작성, 게놈구성분석, 유전자 사이 의 거리를 측정할 수 있는 FISH(Fluorescence in situ hybridization), 종간잡종 등 유전자간 유연관계가 먼 식 물끼리 교배 시 유전자 대합정도를 알 수 있는 GISH (genomic in situ hybridization), 핵 DNA를 분리하여 유전자가 거리가 가까운 유전자사이의 분포여부를 알 수 있는 fibre-FISH, Comparative genomic hybridization 등과 같은 다양한 기법들이 개발되었다. 이로 인해 첫번 째 시대의 한계점이었던 상동염색체의 대합, 염색체의 특 성구명 등이 좀 더 명확히 진행되었다. 또한 분자생물학 의 발달과 함께 DNA-based molecular marker가 개발되 어 염색체 상에서 그 위치를 탐색하여 molecular map의 작성이 이루어졌다. 네번째 시대(1995 ~현재)에는 whole genome sequencing의 진행과 함께 세포유전학적 연구 가 병행되고 있다. Arabidopsis 연구의 경우, genome sequencing project와 함께 다양한 세포유전학적 기술이 적 용개발되어 fiber-FISH(Jackson et al. 1998), chromosome painting(Lysak et al. 2001), FISH mapping(Jackson et al. 2000), BAC-FISH(Ziolkowski and Sadowski 2002) 등의 연구결과를 통해 보고되었다. 국내에서는 genome sequencing project 중 Brassica genome project(Hwang et al. 2009; Lim et al. 2007, 2007; Mun et al., 2011)를 시작으로 하여 Ginseng whole sequencing project(waminal et al. 2012), Radish genome project(Hwang et al. 2012) 등에서 세포유전학적 연구 기법이 이용되었다. 세포유전학 적 연구는 이들 연구에서 염색체지도(chromosomal map)와 유전자지도(genetic map)를 통합하여 통합물리지도(integrated physical map)를 작성하는데 이용이 되고 있다.

    세포유전학 연구의 응용

    화분임성(pollen viability)과 배수성(ploidy level)

    화분의 임성과 배수성은 매우 밀접한 관계가 있으며 효 율적인 교배육종을 위해서는 반드시 확인되어야 하는 기 본요소이다. 배수성을 검경하는 방법은 크게 현미경을 이 용한 관찰법과 배수성 판별기(flow cytometer)를 이용하 는 방법으로 나눌 수 있다. 배수성 판별기를 이용하여 배 수성을 관찰하는 경우, 단시간 안에 많은 시료를 검정할 수 있고 배수성을 확인할 수 있는 재료의 범위(잎, 근단, 줄기, 생장점 등)가 비교적 많은 장점이 있다. 그러나 고 가의 기계 구입이 필요하고 배수성을 확인하게 위해 기 준을 설정하는 과정(control setting)이 제대로 되지 않거 나 국화와 같이 다양한 배수성으로 구성된 식물의 경우 에는 배수성 검경 결과의 재현성과 정확성이 떨어지며 이 수성 식물은 판별이 어려운 단점이 있다. 현미경을 이용 하는 경우는 하나의 세포 안에 있는 염색체의 수(배수성 및 이수성)를 가장 명확하게 관찰할 수 있는 장점이 있 으나 염색체를 관찰하고 분석하기 위한 특별한 기술이 필 요하고 검경까지 걸리는 시간이 길며 염색체를 관찰하기 위해 사용되는 부위가 주로 뿌리나 생장점과 같은 분열 조직으로 한정되는 단점이 있다.

    화분의 임성을 관찰하기 위해서는 염색액을 이용하여 활 력을 측정하거나 인공화분발아배지를 만들어 실제 화분관의 신장을 유도하는 방법(in vitro germination assay)이 적용된 다(Viintin and Bohanec 2004). 주로 사용되는 염색액은 Aceto-carmine(cytoplasmic content), Aniline blue(callose in pollen walls), FDA(fluorescein diacetate, esterase activity), MTT(2,5-diphenyl monotetrazolium bromide, dehydrogenase activity) 등이 있으며 이는 화분효소활성도(pollen enzymatic activity), 세포막의 온전함 정도 등을 측정하여 잠재적으 로 화분관의 신장이 가능한 정상화분과 기형의 비정상화 분을 모양과 염색 정도의 차이를 통해 가시적으로 구별 해 낼 수 있다. 인공발아배지를 통한 측정방법은 인위적 으로 화분관을 신장시키도록 조성된 배지 위에서 화분 중 실제 화분관을 신장하는 화분관 신장율을 측정하는 방법 으로 염색액 검경법 보다는 정확한 화분의 임성율을 측 정할 수 있다. Hwang et al.(2010)은 장미 화분의 임성, 배수성 및 교배조합의 검정한 결과 중 3배체 품종을 모• 부본으로 사용한 교배조합을 조사한 결과, 염색액검 경(Alexander’s solution, Aceto-Carmine)에서는 정상화분 의 비율이 52%로 관찰되었으나 인공발아배지에서는 발 아율이 1%로 정상화분이라 할지라도 실제 발아율은 차 이가 나는 것을 확인하였다. 또한 3배체를 부본으로 하 여 교배결과, 교배 후 결실된 종자에서 모두 발아가 되 지 않아 교배 효율이 떨어졌다. 따라서 교배육성을 하기 위한 모• 부본으로 이용되는 재료는 화분의 조사뿐만 아 니라 염색체의 배수성 검경도 추가적으로 이루어져야 효 율적인 교배육성이 가능할 것으로 생각된다.

    FISH 기법을 이용한 핵형분석

    1960 ~ 70년대에 방사선 동위원소로 표지된 탐침을 탐 색하는 ISH(in situ hybridization) 기법의 이용으로 염색 체 상에서 특정 DNA나 RNA 서열의 위치와 분포가 시 각적으로 확인이 가능하게 되었다(Devi et al. 2005). 그 이후, 탐침 검출 시간의 단축, 해상도의 증가, 비특이적 background의 감소, 화학적으로 장시간 안정한 탐침, 결 과의 재현성 유지 등의 장점을 이용하여 보다 정확한 정 밀핵형분석이 이루어지도록 FISH 기법으로 개선되었다 (Lager-Safe et al. 1982). 광학현미경하에서 관찰이 가능 한 염색액을 주로 이용하는 일반핵형분석의 경우, 염색 체의 크기가 작고 상동염색체의 모양이 비슷하며 동원체 의 위치가 명확히 드러나지 않는 경우 정확한 핵형분석 이 어려움이 있다. 그러나 특정 marker를 사용하는 FISH 핵형분석을 이용하면 보다 명확하게 염색체를 특징 지을 수 있다. FISH 핵형분석을 위해서는 주로 5S와 45S 두 종류의 리보솜 DNA(ribosomal DNA)를 이용하는데 이러한 FISH 핵형분석은 주요 화훼작물 백합(Hwang et al. 2011; Karlov et al. 1999; Lim et al. 2000, 2001; Marasek et al. 2004a, b), 장미(Fernndez-Romero et al. 2001; Hwang et al. 2012; Ma et al. 1997), 국화(Abd El-Twab and Kondo 2003, 2006a, 2006b, 2007, 2008) 등에서 이루 어졌다. 주요 화훼작물의 FISH 핵형분석은 주로 원종을 중심으로 이루어졌으며 이러한 결과는 염색체 수준에서 원 종들의 분류학적 연구에 이용하거나 원종을 교배 모부본 으로 이용하는 교배육성에 기초자료로 이용될 수 있다. 그 러나 원종에 비해 우수 재배종에 대한 염색체 정보는 매 우 미비한 실정으로 우수육성품종에 대한 염색체 정보를 추가적으로 수집할 필요가 있다.

    FISH 기법을 이용한 세포유전학적 연구는 최근에 유전 체 분석 연구와 함께 수행되고 있으며 주로 옥수수, 밀, 토마토, 감자 등과 같은 주요식량작물이나 경제원예작물 에서 많은 연구가 수행되고 있다. 주로 수행되는 연구들 은 genomics research에서부터 연구된 marker등을 이용하 여 heterochromatin이나 euchromatin의 영역을 분석하는 연구(Danilova et al. 2008; Iovene et al. 2008; Koo et al. 2008; Szinay et al. 2008; Tang et al. 2008), 십자화과 (Brassicaceae)(Jackson et al. 2000), 가지과(Solanaceae) (Iovene et al. 2008), 벼과(Poaceae)(Draye et al. 2001)에서의 comparative genomics, chromosomal duplication event 와 같은 evolutionary study(Jackson et al. 2000) 등이 이루어지고 있다. 이에 비해 화훼작물의 연구는 주로 신 품종 육성이나 일부 마커개발(Yuan et al. 2013)이 이루 어지고 있는 실정이다. 앞으로 유용 화훼작물에서도 유 전체 연구가 활발히 이루어지는 것을 예상할 때, 화훼작 물 염색체의 핵형 분석 등의 밑그림을 그려놓는 작업이 필요할 것으로 생각된다.

    GISH 기법을 이용한 alien genome analysis

    Genomic in situ hybridization(GISH) 기법은 유전자의 단편을 이용하는 FISH 기법과는 달리 genomic DNA를 탐침(probe)로 제작하는 기법으로 종속간 교잡식물의 형 성 시 후대의 게놈에 혼합된 서로 다른 양친 게놈 염색 체를 구분하고 유전자의 이입 정도를 확인하는데 매우 유 용한 기법이다(Jacobsen et al. 1995; Schwarzacher et al. 1989; Takahachi et al. 1997). FISH 기법은 ribosomal DNA, single-copy gene, 또는 각종 repeat sequence등의 DNA 단편을 탐침으로 이용하나 GISH 기법은 total genomic DNA 전체를 탐침으로 이용하는 차이가 있다. GISH 기법은 특별히 종간잡종의 감수분열 과정 중에 염 색체 대합(chromosome pairing) 정도를 분석할 수 있으 며 univalent, bivalent, multivalent의 빈도, 재조합이 일 어날 때 chiasma의 형성 정도를 파악할 수 있다. 이러 한 결과는 감수분열 과정에서 종간잡종의 불규칙적인 염 색체 대합상태를 가시적으로 확인할 수 있으며 이는 곧 종간잡종의 임성과 연결되어 설명할 수 있다(Silva and Souza 2013). 또한 GISH 기법을 통해 종간잡종을 통해 서 로 다른 게놈이 유입되었을 때 염색체상에서 translocation 을 모니터링 할 수 있으며 allopolyploid의 origin 게놈을 구 별해 내는데도 응용하여 이용될 수 있다. GISH technique 을 이용한 후대 분석은 화훼류에서는 주로 백합에서 이 루어졌다(Barba-Gonzalez and de los Milagros Revuelta- Arreola 2010; Lim 2000, 2001a, b, 2003; Marasek et al. 2004.

    Microdisscetion 기법을 이용한 유전체 연구

    Microdissection과 microcloning 기법은 Scalenghe et al.(1991)에 의해 개발되었으며 lazer microbeam을 이용 하여 특정 염색체 또는 염색체 단편을 오려내고 PCR 기 술을 이용하여 원하는 유전자를 증폭하는 기술이다. 초 기에는 주로 주요 경제작물(보리, 고추, 귀리, 밀 등)에 적 용되었으며(Albani et al. 1993; Chen and Armstrong 1995; Park and Kim 1999; Sandery et al. 1991; Schondelmaier et al. 1993; Zhou et al. 1999) 화훼류에 서는 참나리 1번 염색체를 microdissection하여 보고되었 다(Hwang and Lim 2011). Microdissection 후 염색체를 template로 하여 cloning 하는 것이 가능한 것은 2가지 PCR 방법이 적용되었기 때문이다. 첫번째 방법인 Linker adaptor mediate PCR(LA-PCR)은 template로 이용되는 chromosomal DNA를 enzyme specific Linker를 부착하고 이 부분을 primer binding site로 이용하여 PCR하는 방법 이다. 두번째 방법인 Degenerate oligonucleotide primed- PCR(DOP-PCR)은 부분적으로 degenerate한 sequence를 가 진 primer를 이용하여 template로 이용하는 whole chromosomal DNA 전체를 target으로 하여 다양한 위치에서 증 폭하는 방법이다. 참나리에 Microdissection 기법을 적용한 결과, 약 2000개의 clone으로 구성된 chromosome specific 한 library를 구축되었다. 이들을 이용하여 sequencing data 분석결과, 백합 특이 반복염기서열(repetitive sequences)을 5 그룹으로 분리할 수 있었으며 이들은 public database에 서는 매치되지 않는 염기서열로 분석되었다. 이와 같은 결 과는 염색체 DNA를 template로 하고 두가지 PCR 방법 에 이용된 primer의 specificity가 떨어지는 것이 백합 1번 염색체 염기서열 중 주로 반복염기서열이 증폭되어 분리된 것으로 생각되며 이를 이용한다면 유전체에서 주 요 반복염기서열을 분석하는데 응용될 수 있을 것으로 생각된다. 또한 백합 특이 반복염기서열 중에서 염색체 1번에서만 관찰되는 염색체 특이마커가 분리되었으며 이 를 이용하여 백합 속의 서로 다른 subsection에 속하는 종에 탐색한 결과 반복염기서열 하에서의 분류학적 마커 로서의 이용 가능이 확인되었다.

    Single gene의 탐색

    FISH 기술의 개발과 함께 multi-copy gene의 detection 뿐만 아니라 low-copy 또는 single gene의 detection 방 법이 요구하게 되었다. 특히 유전자 변형 작물의 도입유 전자 확인은 주로 분자생물학적인 방법으로 확인되었으 나 그 위치의 가시적 확인을 위해 일부 유전자 변형 작 물에 FISH 기법을 이용하여 도입유전자의 염색체 상에 서의 위치 탐색을 시도 하였다. 그러나 multi-copy gene 의 경우 비교적 염색체 상에서의 위치 탐색이 쉬우나 single 또는 low-copy gene의 경우는 탐색이 어려우며(Khrustaleva and Kik 2001) 작은 형광 signal도 sensitive하게 감지 할 수 있는 CCD(cooled charge-coupled device) camera 장치 도 요구한다(Wiegant et al. 1991). 또한 식물세포는 cell wall의 물리적 장벽 때문에 동물세포에 비해 작은 크기 의 gene의 탐색이 비교적 어렵다. 이를 보완하기 위해 Tyr-FISH 기법의 개발 등 여러 연구자들에 의해 single gene을 detection하기 위한 연구가 이루어졌다(Kato et al. 2006; Khrustaleva and Kik 2001; Wang et al. 2006; Yu et al. 2007). 이를 통해 국외에서는 양파에서 ultrasensitive한 FISH 기법이 적용되어 710bp의 uidA gene의 위치가 보고되었다(Khrustaleva and Kik 2001). 국내에서 는 CMVPO-CP 도입 GM 고추의 도입유전자 위치(Lee et al. 2011)와 single trait이 도입된 Cry1Ac1과 LS28 line의 벼에서 도입 유전자의 위치가 확인되었으며 GM 벼 event 간에 교배가 된 stacted GM event상에서 stack gene의 위치 확인 연구가 이루어졌다(Park et al. 2010). 화훼류 에서는 GM 페튜니아에서 1.4kb의 single-copy chalcone synthase A (chsA) gene이 한쌍의 염색체 위에서 탐색되 어 보고되었다.

    이러한 유전자 변형 작물 도입유전자의 염색체 내에서 의 탐색 연구는 도입유전자의 안정적 삽입여부 확인(위 치 및 copy 수), 유전자 변형 event 간에 교배가 된 stacked GM event상에서 stack gene의 염색체에서의 위 치, 유전자 변형 작물의 gene flow 탐색 등에 다양하게 응용될 수 있다. 뿐만 아니라 전세계적으로 유전자 변형 작물의 개발과 상업화가 급격히 증가 추세에 있어 유전 자 변형 작물의 생산 후 염색체의 수적, 구조적 안정성 을 지속적으로 모니터링하는 작업도 반드시 요구된다.

    초 록

    세포유전학 연구는 유전학과 세포학으로부터 발생된 학 문으로 염색체의 수, 구조, 행동 등의 염색체 연구를 기 본으로 하는 분야이다. 1980년대 이후 분자생물학적 기 술의 적용으로 새로이 발전하였으며 현재는 유전체 해 독 연구와 함께 다양한 연구분야에 적용되고 있다. 세 포유전학적 연구를 위한 연구기법은 전통염색법에 의한 banding 기법, fluorescence in situ hybridization(FISH), genomic in situ hybridization(GISH), fiber FISH, BAC-FISH 기법 등이 있으며 이들은 기초 핵형을 분석 하는 연구 분야에서부터 유전자의 위치를 탐색하는 응용 연구분야까지 다양하게 이용되고 있다. 현재 화훼연구에 있어서 세포유전학적 연구의 이용은 (1) 배수성 검경을 통한 교배육성의 기초자료 제공, (2) FISH 핵형분석을 통 한 정밀한 핵형분석, (3) GISH 기법을 이용한 게놈 이 입분석, (4) microdissection 방법을 이용한 유전체 분석, (5) GM 작물에서 외래유전자의 위치 추적 등에 적용될 수 있다. 그러나 국내의 화훼연구에서의 세포유전학적 연 구는 아직은 크게 활성화 되지 않았으며 추후 유전체분 석연구 응용을 위해서는 다양한 작물과 다양한 기술의 적 용이 필요하다. 따라서 본 논문에서는 화훼연구에서의 세 포유전학의 개념과 필요성, 연구의 이용에 대해 논하고 자 한다.
